Valdoxan (Agomelatine)

Agomelatine - agonist melatonin (MT1 and MT2 receptors)and the antagonist5HT2c.Represents an energizer, active on validate depression models (the test of the learnt feebleness, the despair test, chronic stress of moderate expressiveness), no less than at models with decinhronise a warm rhythm, and also in experimental situations of alarm and stress. In experiments on animals with the simulated syndrome of the braked phase of a dream (Delayed Sleep Phase Syndrome) at blind and old animals it has been shown that Valdoxan restores synchronisation cirkad rhythms through stimulation melatonine receptors. At chronic stress Valdoxan interferes with occurrence "broken off" (fragmented) a dream. In experiences on healthy volunteers Agomelatine did not break normal structure of a dream and well influenced a dream at sick of depression. In therapeutic doses Valdoxan prevented development of a sleeplessness and infringements of memory from the moment of reception of a preparation and till the morning.

Agomelatine does not cause accustoming that has been shown in research on healthy volunteers with use of a visual analogue scale or the Questionnaire of the Research Center on studying of fatal predilections (Addiction Research Center Inventory - ARCI 49 check-list). Valdoxan also investigated about occurrence of a syndrome of cancellation at sick of depression by means of a Questionnaire revealing signs of a syndrome of cancellation (Discontinuation Emergent Signs and Symptoms (DESS)). It was found out that the cancellation syndrome does not develop even at the sharp termination of treatment.

During the program of clinical development Valdoxan its efficiency and safety against the big depressive frustration was investigated. Researches with the active control have confirmed the received results.

Valdoxan also it was effective at initially high levels of alarm, no less than at a combination of disturbing and depressive frustration. Influence Agomelatine on sexual function sick of depression with remmitic a clinical course was studied. The Scale of studying of influence on sexual sphere (Sex Effects Scale - SEXFX) was used. It has been shown that Agomelatine does not cause sexual infringements and does not influence occurrence of excitation and orgazm. At sick of depression, since second week of treatment, Valdoxan statistically signi/ficantly improved process sleep, without causing thus the subsequent day block.

  5. its a pretty good medicine for depression and sleep but its not true that there are no gasto-intestinal problems, many people report stomachcramps and pain. its because theres a burning sensation to the tablet that really upsets the stomach with a lot of people.

  8. I feel better than I did before taking it for depression and anxiety. But have had the most vivid dreams. After a night out and getting drunk, I had nightmares that were very real. I woke up thinking the 86 y/o lady downstairs had got into my flat and was coming to stab me with a knife. I have also had lovely dreams where people care for me and I felt very loved.
    Also I feel energetic when I'm awake a lot and want to get things done and have to calm myself down not to go too fast and make mistakes.
    I find myself talking and telling everyone everything. (slight exaggeration there, but not much)

  9. I tried this drug for 2 months. I was so positive and excited about it - based on the way it's supposed to work, I thought it would be perfect for me. It had absolutely no effect whatsoever, except to relieve my wallet of €140. Now that's depressing! :-(

  10. My wife has bipolar and after trying many AD’s to supplement lithium was put on Valdoxan 25mg by the psychiatrist. She is having a very serious reaction to it. Was stopped after 2 doses but 5 days later she is still in severe pain (she says it is worse than giving birth). The pain is back and stomach. She is also confused with poor balance, unable to write etc and is generally pretty incapacitated.
    The psychiatrist has done basic tests – pulse, blood pressure, (both OK) blood test for lithium (result awaited, but he thought it unlikely to be the cause). He has done a severe reaction report. Currently she is taking pain killers + valium + sometimes alcohol to deal with the pain – this puts her out for a while. The drug will have long gone from her system so the worry now is has it done permanent damage. I’ve asked him to think of a plan B if there is no improvement soon.

    If this is gastritis does anyone have
    experience of how long it takes to stop?

  11. I was so positive, I thought it would be ideal for me ! It didn't work, I have yet to meet anyone it works for. I went anxious and got very bad stomach cramps.

I believe the stomacke pain is related to the change in daily rythm of your digestive system, and that the messing with the melatonin ruled reactions in your stomack/liver/other parts of your digestive system combined with the forced changes on the serotonins widespread regulating of your digestive system , taht all this in combination can make longlasting disturbances in your inner machinery, but that after a while it will regulate back to more normal painless ditgesting...

    the longer you continue taking the evil pill the longer and more the severe affect will be and in a more and more cronical feature

Yes I had body changes too, tummy problems but they managed me while in hospital, but coming from another drug for depression to this one after two weeks of adjustment my family said I was brighter, I actually got dressed and got up and around and do so a bit more often today. I guess each to their own on this. Sadly my dose just increased but I have a lot of other medical problems too much to talk about here..

  14. Sleep awesome, wake up renewed and refreshed and a new lease on life. Horrible digestive issues and my ass feels like it's had a firework shoved up it or I've been raped with a barbwire condom. Guts make it hard to function but atleast the desire to do so is there.
